It's possible, but it would be a lot of work and I cannot see the point. Why do you think you need this functionality?

The reason why we stick to the square base size is that it simplify things. For example we have to draw the building graphic only once. If it wouldn't be a square we would have to split it up into parts and draw them in the correct order.

Instead of creating a single 2x4 building you may create two 2x2 buildings that fit together. The result will be the same

Yea, if 2x4 building rotates, it becomes 4x2, and vice versa
